DNA Sequencing Kits
Oligonucleotide sequencing by pyrosequencing uses a single-stranded template of DNA and is a common step in cloning experiments. It involves synthesizing the complementary strand of DNA and the release of light by luciferase, after the addition of each complementary nucleotide. These kits are limited to around 500 nucleotides, so will not be useful if you are looking to sequence longer strands of DNA.
Sanger Sequencing kits on the other hand, use selective chain termination during in vitro replication; adding modified dideoxynucleoside triphosphates (ddNTPs) to cells which terminate elongation of the complementary strand. The fragments of DNA created, can then be used to identify the sequence when ran on a polyacrylamide gel. Sanger sequencing kits generally have a longer nucleotide maximal limit, at around 800-1000 nucleotides. This technique has mostly been replaced by Next-Generation Sequencing (NGS).
Whole-genome sequencing constructs a complete analysis of an entire genome, or identifies mutations, as an alternative to genome mapping. With the advent of Next-Generation Sequencing (NGS), and with NGS kits on the market, this technique is becoming more efficient and easier to analyze.
As for other experiments, the exact type of DNA and sample source is an important factor to consider. Be sure to have a look at our DNA sequencing kits to find the right one for you. There are kits to specifically match the DNA you are sequencing, including kits for; ssDNA, plasmid DNA and genomic DNA.
Check out our Sanger Sequencing Troubleshoot for more guidance on this technique.
RNA Sequencing Kits
RNA sequencing (whole transcriptome shotgun sequencing) is a gene expression analysis technique that employs NGS. It allows for the analysis of low-expressing genes as well as alternative splicing or post transcriptional RNA modification. RNA sequencing can also be applied to miRNA or other non-coding RNAs, creating a powerful tool for RNA research. There are dedicated kits for each RNA target on the market, ready to make these experiments easy for you.
Our Next Generation Sequencing (NGS) Troubleshoot offers advice on recurrent NGS problems.
Compare DNA Sequencing Kits
With our compare function, you can avoid all the time and energy wasted sifting through multiple web pages from different suppliers. At ZAGENO you can clearly see kits side-by-side, with the relevant attributes for each kit neatly in line for easy selection of the best product for you.
Click on the comparison below for a clearer view!
For example, this comparison shows:
that the BIGDYE® TERMINATOR V3.1 CYCLE SEQUENCING Kit from Applied Biosystems accepts a wide range of starting material.
that the PYROMARK PCR Kit from Qiagen is the cheapest per reaction.
that the SEQPLEX DNA AMPLIFICATION Kit from Sigma-Aldrich can use less than 1 ng of sample as a result of NGS.
This comparison clearly exemplifies how every kit has strengths and weaknesses. Depending on what features you require for your experiment, these details should enable you to make an informed decision on the right kit for you.
The ZAGENO comparison does not highlight one kit to be better than the other, as the kit of choice may vary between researchers - depending on each individual's preferred attributes. The best kit is the one that meets your needs - ZAGENO allows you to make an informed decision with minimum effort.
