Next generation sequencing

For Next Generation Sequencing there are many parameters to consider when creating a library. Once you know what kind of library you need, consider the size of the fragments. Short fragments can be amplified more efficiently while longer ones create better, more diffused sequencing clusters. The length of fragments might also be dictated by the nature of the template, the genome of interest, and the characteristics of its transcript.

There are also a few variables to think about for fragment creation. Physical DNA shearing methods such as sonication generally create fewer artifacts than enzymatic ones. But the latter are typically faster, reducing the overall sample preparation time.

RNA sequencing

For RNA sequencing applications, there are dedicated cDNA library kits available. Some kits maintain the original orientation of RNA in the cDNA copies,which is required for some applications. Others include only cDNA, avoiding DNA amplification from other RNA species, or to solve complex secondary RNA structures. Thus each kit can be tailored to fit your experimental parameters.
