Description

VCAM-1, which is present on activated endothelial cells. VCAM-1 is an adhesion molecule, which binds to the integrin alpha4/beta1 (VLA-4) and to the integrin alpha4/beta7 on lymphocytes. This antibody has been shown to inhibit T lymphocyte adhesion to TNF stimulated endothelial cells. The molecular weight of the recognized antigen is 110kD.Applications:Suitable for use in ELISA and Western Blot. Other applications not tested.Recommended Dilution:ELISA: To detect hVCAM-1 by sandwich ELISA (using 100ul/well antibody solution) a concentration of 0.5-2ug/ml of this antibody is required. This antigen affinity purified antibody, in conjunction with Biotinylated Anti-Human VCAM-1 as a detection antibody, allows the detection of at least 0.2-0.4ng/well of recombinant hVCAM-1.Western Blot: To detect hVCAM-1 by Western Blot analysis this antibody can be used at a concentration of 0.1-0.2ug/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant hVCAM-1 is 1.5-3ng/lane, under either reducing or non-reducing conditions.Optimal dilutions to be determined by the researcher.Storage and Stability:Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
