Description

Klotho is a type-I membrane protein that is related to beta-glucosidases. Reduced production of this protein has been observed in patients with chronic renal failure (CRF), and this may be one of the factors underlying the degenerative processes (e.g., arteriosclerosis, osteoporosis, and skin atrophy) seen in CRF. Also, mutations within this protein have been associated with ageing and bone loss.Applications:Suitable for use in ELISA and Western Blot. Other applications not tested.Recommended Dilution:ELISA: To detect hKlotho by sandwich ELISA (using 100ul/well antibody solution) a concentration of 0.25-1ug/ml of this antibody is required. This biotinylated polyclonal antibody, in conjunction with Polyclonal Anti-Human Klotho as a capture antibody, allows the detection of at least 0.2-0.4ng/well of recombinant hKlotho.ELISA (Direct): To detect hKlotho by direct ELISA (using 100ul/well antibody solution) a concentration of 0.25-1ug/ml of this antibody is required. This biotinylated polyclonal antibody, in conjunction with compatible secondary reagents, allows the detection of at least 0.2-0.4ng/well of recombinant hKlotho.Western Blot: To detect hKlotho by Western Blot analysis this antibody can be used at a concentration of 0.1-0.2ug/ml. Used in conjunction with compatible secondary reagents the detection limit for recombinant hKlotho is 1.5- 3ng/lane, under either reducing or non-reducing conditions.Optimal dilutions to be determined by the researcher.Storage and Stability:Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
